n. 1. A dependence on a drug in which the person feels pleasure or release from tension in using the substance and fears being without the drug. 2. The process of altering metabolism to accommodate the continuing presence of a drug in the body so that the person needs increasing doses to reach a given level of reaction (tolerance) and unpleasant and sometimes dangerous consequences when the substance is withdrawn.
